It is possible to count the number of plant cells in a measured volume of water. Because nets are not fine enough to filter the smallest plants, the phytoplankton is usually obtained by collecting water samples and removing the cells by centrifuging or sedimentation. Subsamples are made with a haemocytometer and counted under a microscope. Alternatively, a Coulter counter can be adapted for counting and recording numbers of small planktonts. This electrical instrument responds to changes in an electrical field as small particles in an electrolyte pass through an aperture. When the number of plant cells in unit volume of water has been determined, the weight of plant protoplasm must be calculated. For this it is necessary to know the size and weight of the plant cells, and to make due allowance for the inorganic content of the cells.
